I have been trying to dry my 4-12% gradient polyacrylamide gel without success. I have tried soaking for 20 minutes in just deionized h20, various concentrations of methanol and glycerol, different gel dryers. We are using a vacuum gel drying system and we are able to get a gel to dry without cracking but only if the gel is an 8% gel.

1. Soak the gel in a solution of 30% methanol, 3% glycerol for 15-30 minutes. This will help minimize gel cracking during drying.
Acrylamide is a building block for the polymer, polyacrylamide, a material well-known in molecular biology laboratories as a gel matrix for resolving DNA fragments in sequence analysis and identifying proteins, both under electric fields. In the world at large, polyacrylamide is used in water purification to flocculate suspended organic matter.

Why do my pictures of large sequencing polyacrylamide gels come out blurry? ... I think u can fix the gel in 20-30% methanol..this will give u sharper bands.. ... not patchy. The best option is ...
Set the voltage upto 180 V and run for 1 hour.(Don't allow the dye front to go out of the gel). Staining the gel: After running, switch off the power supply and take out the gel plates, remove the gel. Place the gel in the staining solution for 30 minutes. Destain the gel until the bands are properly seen.

Introduction to Agarose and Polyacrylamide Gel Electrophoresis Matrices with Respect to Their Detection Sensitivities 5 greater resolving power, can accommodate larger quantities of DNA without significant loss in resolution and the DNA recovered from polyacrylamide gels is extremely pure (Guilliatt, 2002).
Polyacrylamide gel electrophoresis (PAGE) is a technique widely used in biochemistry, forensic chemistry, genetics, molecular biology and biotechnology to separate biological macromolecules, usually proteins or nucleic acids, according to their electrophoretic mobility.Electrophoretic mobility is a function of the length, conformation and charge of the molecule.
